Outline of Annual Research Achievements

本研究では、リン酸緩衝生理食塩水（PBS）と脱イオン蒸留水（DDW）に対して定量化された酸素ラジカルを照射し、各溶液に生成された過酸化水素、次亜塩素酸、亜硝酸・硝酸イオン濃度などを定量し、液中での反応メカニズムを解明した。その結果、酸素ラジカルにより生成されたOHラジカルがPBS中の塩素イオンと反応し、次亜塩素酸を生成されることが判明した。1分間の酸素ラジカル照射で生成された次亜塩素酸により大腸菌は滅菌されるが、酵母は1桁程度の不活性化にとどまり次亜塩素酸に対する耐性が高いことが分かった。さらに30秒程度（酸素原子のドーズ量1×1017cm-3）の短時間照射の場合、酵母は殺菌ではなく20％程度増殖が促進されることが分かった。また、酸化窒素ラジカルを選択的に照射した実験ではNOのドーズ量が1×1019㎝-3の時20％程度の増殖促進が見られ、酸素ラジカル照射とは異なりPBS溶液中だけでなくDDW中でも同様な増殖促進が見られることが分かった。これらの結果から増殖促進には次亜塩素酸ではなく他の活性種が寄与している可能性が明らかとなった。さらに培養細胞である胎児マウスの線維芽細胞に対しても20％以上の増殖促進が見られることが判明し、皮膚細胞の増殖時間を短縮できる可能性を得ることに成功した。また、酸素ラジカル照射した培養液をメラノーマ細胞に作用させ不活性化されるシグナルパスを検証した結果、酸化ストレスによりミトコンドリアの膜電位が下がり、カスパーゼ９と３が活性化されることでアポトーシスが誘導され不活性化されているということが明らかとなった。
