| Outline of Annual Research Achievements |
本申請課題では、骨減少性疾患において有用であると考えられる、「骨吸収を抑制すると同時に骨形成を促進する天然物」の作用を、細胞および個体レベルで解明するために研究を進め、これまでの研究で、卵巣摘出骨粗鬆症モデルにおいて、honokiol
の腹腔投与が骨稜の減少を結いに抑制することを明らかにしている。また、honokiolの骨芽細胞分化促進作用に核内受容体のRXRが関わっている可能性とShhシグナルが関わっている可能性を見出している。
平成28年度は候補化合物であるharmineおよびhonokiolが共通して相互作用する生体分子として、ABCトランスポーターの一つのABCB1に着目して、骨代謝細胞の分化調節作用とABCB1の関連を明らかにするために、ABCB1と相互作用する化合物の探索を行った。その結果、南米原産植物Pterogynine Nitensに含まれるNitensidine Aと、生薬シンイに含まれることが知られるEpimagnolin AがABCB1と相互作用することを見出した。
さらに、ドッキングシミュレーション解析によって、ABCB1とNitensidine AおよびEpimagnolin Aの相互作用部位について検討し、それぞれの結合に重要なアミノ酸残基候補を同定した。これらの化合物と骨代謝との関わりを明らかにすることで、骨代謝におけるABCB1の関与とHonokiolおよびHarmineの作用機序への関与が明らかになることが期待される。
