| Outline of Annual Research Achievements |
環境による健康問題として、PM2.5（微小粒子状物質）などによる環境中の粒子に加え、ナノ素材などの新規工業的素材の健康への影響に社会的関心が高まり、生体影響に関するリスク評価や環境基準値の設定が求められている。本研究では、ナノ素材の血管系に対する影響とその障害機構を解明し、外部環境ストレスによる血管系に対するリスク評価のための科学的基礎資料を作成することを目的とする。
本年度は、血管内皮細胞を培養し、単層カーボンナノチューブと多層カーボンナノチューブを投与し、MTS assay法にて細胞生存率を検討した。カーボンナノチューブ投与後の細胞上清で、細胞遊走に関与する単球走化性タンパク質の活性を分析し、カーボンナノチューブ投与後の血管内皮細胞における接着因子の発現をウェスタンブロット法で解析した。また、動脈硬化モデル動物であるApoEノックアウトマウスに、単層カーボンナノチューブと多層カーボンナノチューブを投与したところ、動脈硬化の促進が認められることを大動脈弓部のoil red染色で確認した。更に、動脈硬化の発症において重要な役割を果たす接着因子の発現の亢進がその作用機序の一部であることを明らかにした。
