| Outline of Annual Research Achievements |
骨肉腫の新規治療法として，Glycogen synthase kinase 3β（GSK-3β）阻害薬の有用性について検討した．GSK-3βはグリコーゲン代謝に関わる酵素であり，前立腺癌，大腸癌や膵癌などいくつかのがん細胞増殖に関与することがわかっている．また，Wnt/β-catenin経路やNF-κB経路にも関与するといわれている．本研究では，骨肉腫におけるGSK-3βの発現，GSK-3βを阻害することによる腫瘍への影響を観察することにより，治療標的としてのGSK-3βの可能性を検討した．ヒト骨肉腫細胞株HOS，MG63，143-B，Saos-2を用い，コントロールとしてヒト骨芽細胞株hFOB 1.19におけるGSK-3βの発現をWestern blottingで比較したところ，骨肉腫細胞株において優位に高い発現が見られた．また，これらの細胞株をGSK-3β阻害薬AR-A014418，SB-216763で刺激したところ，骨肉腫細胞の増殖を有意に低下させた．また，Tunel imaging assayにおいて，GSK-3β阻害薬は骨肉腫細胞のアポトーシスを誘導し，骨芽細胞においてはアポトーシスに影響を示さなかった．つぎに，ヌードマウスの脛骨に143-B細胞を移植することにより骨肉腫の同所性モデルを作成し，GSK-3β阻害薬（AR-A014418，SB-216763）の腫瘍抑制効果を評価した．腫瘍体積の評価において，GSK-3β阻害薬は治療開始から2週以降で有意な腫瘍増殖抑制を示し，切除した腫瘍重量においても有意な抑制効果を認めた．また，マウスにおいて明らかな副作用は見られなかった．本研究の結果から，GSK-3βは骨肉腫の新規治療標的となる可能性が示唆された．
