Outline of Annual Research Achievements

In the H26 Fiscal year, we were successful in accomplishing all research goals, plus collection of grey langur samples for comparative purposes based from mid-term evaluation of the data set. Two trips were made by MAH and one trip accompanied by YK to Sri Lanka for the collection of fecal samples from toque macaques, purple faced langurs. For comparative purposes, additional fecal samples were collected from grey langurs. In H26, a total of 50 fecal samples from these three species (toque macaque= 11 individuals.; purple face langur= 3 individuals; grey langur= 36 individuals) from were collected from 14 different populations across the distribution of these three species and their respective sub-species.

The fecal DNA was extracted at the University of Sri Jayawardenepura together with on-site collaborator Charmalie Nahallage and a local student assistant. The successfully amplified PCR product was sequenced back in Japan in our laboratory. Our results of the toque macaque were consistent with our previous studies showing that the three putative sub-species, based on morphological traits, sorted into only two haplotype clusters (A and B). Purple face langur sequencing is still underway, but grey langur sequences sorted out into one major cluster as expected by morphological taxonomy, but surprisingly an additional 4 sub-clusters were recognized in populations co-existing with purple face langurs. Further sample collection will be necessary before coming to firm conclusions but purple face langurs and grey langurs sometimes clustered into the same haplotype.

Reason

We were able to locate and collect samples from more groups then previous trips. We were able to conduct the work with the assistance of locally hired students and our local collaborator to collect some samples for us during periods outside of our visits.

Strategy for Future Research Activity

Based on the results thus far obtained and from ongoing DNA sequencing of the purple face langur, we will expand our collection of samples from previously collected sites for toque macaques, grey langurs and purple face langurs, in particular where they are found together in contact zones. Collection is now underway at two field sites in verified contact zones to gain a better perspective of site specific tendencies. Next work will also be expanded to design primers to ID key diagnostic parasite species of these primates.
