Outline of Annual Research Achievements

細胞内寄生体であるトキソプラズマに感染すると免疫正常者では脳、眼、筋肉等にシストを形成するのみで、ほとんど無症状であるが、宿主が免疫抑制状態に陥ると重篤な症状を引き起こすことが知られている。眼は免疫特権部位であり、網膜の微細な病変でも視機能に重大な影響を及ぼし特に小児でのブドウ膜炎の原因としてトキソプラズマ性網脈絡膜炎が第一位となっている。しかしながら、日和見感染症であることから、確定診断や病態把握が困難である。
今回の研究の目的は、トキソプラズマ性網脈絡膜炎発症に関与する蛋白質を網羅的に解析することにより微量検体量でかつ高精度で診断できる新規のバイオマーカーの開発や分子標的治療に資する蛋白質を探索することである。
一年目はトキソプラズマをトキソプラズマ感受性マウスに感染させ２週間後及び４週間後に眼底カメラで眼底を観察し感染成立の有無を確認し眼球摘出後網膜を剥離し蛋白質を抽出し２次元電気泳動を行い、画像解析の結果多数の蛋白質スポットを得た。
二年目は一年目で得られた蛋白質スポットの内、感染前後で発現の差が２倍以上の蛋白質を切り出し、ゲル内消化を行い、液体クロマトグラフ・タンデム型質量分析計（ＬＣ－ＭＳ／ＭＳ）で測定しデータベース検索の結果、感染後に２倍以上上昇した８種類、２倍以上減少した４５種類の蛋白質を同定した。
また、前房水及び硝子体中のバイオマーカーを探索するべく、感染前後の前房水及び硝子体液、血清、髄液を約１００匹のマウスから採取し凍結させており、今後、各種サイトカイン、ケモカイン等の濃度を測定する予定である。

Reason

マウスの眼球から前房水や硝子体液、またマウス脊髄腔から髄液を採取するのに技術的な問題があり、時間がかかっている。これらの体液は一匹のマウスから数マイクロリッターしか採取できず、解析に必要な量を得るのに各群約１００匹のマウスが必要であることも進捗が遅れている理由となっている。

Strategy for Future Research Activity

トキソプラズマ感染前後のマウス（各々約１００匹）から前房水、髄液、血清をすでに凍結保存してあるので、各種サイトカイン、ケモカイン等の解析を行う。

Causes of Carryover

購入試薬の値段が予定より安価であったため、当初計画で割り当てていた使用額との差額が生じた。
