Outline of Annual Research Achievements

昨年度までに同定したPB2分節を由来とする新規ウイルス蛋白質PB2-S1について、A型インフルエンザウイルスの感染宿主を鳥類から哺乳動物へ変化させたときに、発現パターンがどのように変化するのかを解析した。
PB2-S1蛋白質は、ヒト由来のA549細胞、マウス由来のL929細胞およびイヌ由来のMDCK細胞では発現されていた。一方、ニワトリ由来のDF-1細胞にウイルスを感染させて、PB2-S1蛋白質の発現を検出したところ、PB2-S1の発現を検出することはできなかった。このことは、PB2-S1の発現が動物種特異的に制御されていることを示唆する結果である。一方で、鳥由来のA型インフルエンザウイルスのPB2-S1の発現を解析したところ、鳥由来のインフルエンザウイルスは、MDCK細胞においてPB2-S1蛋白質を発現しないことが分かった。
以上の結果から、A型インフルエンザウイルスがトリからヒトへと種の壁を越えて感染を拡大する際に、PB2-S1蛋白質がなにか一定の役割を果たしていた可能性を示唆している。種の壁を越えたA型インフルエンザウイルスがヒトの世界に定着した後もPB2-S1蛋白質の発現が80年に渡り維持されていたことから、PB2-S1蛋白質がA型インフルエンザウイルスのヒトからヒトへの感染においても、必要だったことが考えられる。しかしながら、PB2-S1蛋白質の発現を失くしたウイルスのマウスにおける病原性は、PB2-S1蛋白質を発現するウイルスと同様であったことから、その機能は未知のままである。
