| Outline of Annual Research Achievements |
本研究の目的は、世界で初めて新生児感染症において次世代シークエンサーを用いたメタゲノミック診断法を確立し、今まで同定が困難だった病原微生物の同定を行うことである。そのために、まず本研究課題内容を神戸大学大学院医学研究科遺伝子解析研究倫理審査委員会に申請した。No.104「次世代シークエンサーを用いたメタゲノミック診断法の確立と周産期・新生児期・小児期における感染症の新規病原微生物遺伝子の同定」として承認を得た（平成26年9月29日付）。本年度は、1. 研究のセットアップと2. すでに病原微生物を同定できている感染症を発症した既存サンプルを用いて、次世代シークエンサーによるメタゲノミック解析（後ろ向き解析）を行うことを目的とした。
1.Miseq（イルミナ社）プラットフォーム、シークエンスに必要なライブラリ作成技術・シークエンスランや解析を行う研究環境を整えた。
2.エンテロウイルスが同定されている既存サンプル（咽頭ぬぐい液、糞便）で、ライブラリ作成、シークエンスランを行い、ヒトゲノム参照配列を除外し、サンガー法でエンテロウイルスの配列を確認した。
周産期・新生児感染症関連成果の学会発表、論文や著書を執筆掲載した。

本研究課題を平成26年9月29日付で神戸大学大学院医学研究科遺伝子解析研究倫理審査委員会の承認を得た。Miseqプラットフォーム、シークエンスに必要なライブラリ作成技術・シークエンスランや解析を行う研究環境のセットアップを終えた。すでに病原微生物（エンテロウイルス）を同定できている既存サンプル（咽頭ぬぐい液、糞便）で、ライブラリ作成、シークエンスランを行い、ヒトゲノム参照配列を除外し、サンガー法でエンテロウイルスの配列を確認できた。周産期・新生児感染症関連成果の学会発表、論文や著書を執筆掲載した。

| Strategy for Future Research Activity |
既知および未知の病原微生物を検出するための次世代シークエンサーを用いたメタゲノミック診断法の確立の確立を目指す。エンテロウイルスやその他の病原微生物を同定できている既存サンプルを用いてメタゲノミック解析を反復して行い、本診断方法をまず確立させる。続いて、従来法にて病原微生物を同定できなかったが感染症を発症した新生児の既存サンプルを用いて、今まで同定が困難だった既知の病原微生物や新規の病原微生物の同定を試みる。共同研究として藤田保健衛生大学総合医科学研究所分子遺伝学教室倉橋浩樹教授との共同研究でバイオインフォマティクスのアドバイスも受け、衛生研究所との共同研究によりウイルス分離や既知病原微生物の除外診断が可能であるため試料の提出の準備を整えていく。

他研究費により物品試薬費用を充当できた。そのため若干の次年度使用額が生じた。

引き続き、Miseqおよびシークエンスに必要なライブラリ作成・シークエンスランや解析のための試薬等に充当する。積極的に学会へ参加し最新の技術や知識の習得のために使用する。また、成果発表費用としても使用する予定である。
