Research Abstract

A high frequency synchronous somatic embryogenesis from single cells was estalished in carrot suspension cultures. The following results were obtained using this system.
1. Polarities of cell division activity, DNA and RNA synthesis, mRNA and calcium localization were observed during the process from single cells to embryogenic cell clusters. The apperance of these polarities are closely associated with embryogenesis, suggesting that it may play important roles in determination of somatic embryogenesis.
2. The technique of microinjection to single cells was established. Seventy % of microinjected cells could divide and 50 % could differentiate to embryos. DNA fragments from Ti plasmid as a foreign DNA was microinjected to single cells and it was confirmed that introduced foreign DNA was replicated and expressed after differentiation to embryos at 5 % frequency of transformation.
3. Specific protein for embryogenesis was detected. Microinjection of the monoclonal antibody for the protein to single cells inhibited embryogenesis, indicating that the protein may play a critical role in embryogenesis.
