細胞質ダイニンのアイドリング複合体の構造解析と活性化メカニズムの解明

Research Abstract

我々は細胞質ダイニンの制御機構の解明に取り組んできた。細胞質ダイニンは微小管上を双方向に走るがプラス端に向って走るメカニズムは不明であった。我々は滑脳症の原因遺伝子・LIS1が細胞質ダイニンを微小管上に固定し、微小管-LIS1-細胞質ダイニンの複合体を形成し、キネシン依存的に運搬することを明らかにした。さらにアスペルギルスにおけるNud遺伝子群のNudCがキネシンのアダプタータンパク質として機能していることを明らかにした。このことからLIS1が変異を起こすと細胞質ダイニンの順行性の運搬が障害され、細胞内における細胞質ダイニンの局在が中心体に偏った分布を示し、細胞の末梢部分で枯渇することが分かった。このことがLIS1の変異に伴う神経細胞の遊走異常、また細胞分裂における紡錘体形成や染色体分配の異常につながることが分かってきた。また、NudCの変異は細胞質ダイニンの順行性の運搬のみならず、他のオルガネラの運搬も障害されることから、NudCはキネシンの一般的なアダプタータンパク質として機能していることが示唆された。さらに、細胞質ダイニンはLIS1により微小管上にアイドリング状態になるが、低分子量G蛋白質のRabファミリーのタンパク質によって活性化されることが示唆された。
