脊髄損傷後機能代償回路の特定と新規回復促進戦略の開発

Research Abstract

3頭のマカクザルに上肢の到達-精密把持運動を訓練し、麻酔下にて頸髄C6から胸髄Th1髄節の間の前角に1ミリ間隔で高頻度逆行性レンチウィルスにテトラサイクリン反応性配列TREと緑色蛍光タンパクGFPと破傷風毒素(eTeNT)を搭載したもの(HiRet-TRE-eGFP.eTeNT)を注入、そして1週間後に頸髄C2-C5にかけての中間帯に高感度Tet-ON配列rtTAV16を搭載したアデノ随伴ウィルスベクター(AAV2-CMV-rtTAV16)を注入した。これにより、頸髄C2-C5髄節に細胞体があり、軸索を下部頸髄の運動神経核に投射する脊髄固有ニューロン(PN)が、ドキシサイクリン(Dox)投与中に伝達を遮断されることになる。このようにして、1頭のサルではPNを遮断しながらC5で皮質脊髄路を遮断したところ、通常であれば1-2か月以内に精密把持の回復がみられるところが、4か月経過しても回復は不全だった。一方で、2頭については、C5での皮質脊髄路を切断し、訓練によって精密把持がほぼ回復したところでDoxを投与してPNを遮断したところ、精密恥運動に部分的障害が観察された。これらの結果から、C5での皮質脊髄路の損傷後の機能回復にPNが必要であることが明らかになった。
また、内包出欠モデルのラットの上肢運動の回復過程に、皮質ー赤核路が重要な役割を果たしているらしいことが明らかになってきたので、この経路を上述のウィルスベクター2重感染系を用いて遮断されたところ、回復していた上肢運動が再度傷害された。このような結果から上肢機能の回復に皮質ー赤核路が関与することが証明された。

Reason

マカクザルを用いて、皮質脊髄路損傷後の機能回復過程に脊髄固有ニューロンが関与していることを証明できた。また、ラットで内包出血モデルでの機能回復過程に皮質ー赤核路が重要な役割を果たしていることが明らかになった。今後、これら重要な中継ニューロンに成長因子を発現させる実験が平成26年度大きく発展すると期待される。

Strategy for Future Research Activity

まずはラットのモデルを用いて機能回復にかかわる経路に上記のウィルスベクター2重感染系を用いて成長因子を発現させ、機能回復が促進されるかを観察する。うまくいけば、次にマカクザルを用いて同様な実験を行い、機能代償機構を検証する。
