Outline of Research at the Start

受精は、精子と卵子が一つとなり、新しい命がスタートするドラマティックなステージである。この受精を通じて精子と卵子ゲノムは再プログラム化を受け、胎盤を含めたあらゆる組織へ分化する能力、すなわち全能性を獲得する。本領域「全能性プログラム」では、全能性のメカニズムの分子基盤を解明するとともに、その制御に挑戦する。総括班は、各研究課題を有機的かつ緊密に連携させ、相乗的に研究を推進させる。その成果は論文、プレス発表、シンポジウム、ホームページ等を通じて社会へ発信する。5年後には、本領域を学際的な「全能性研究」ハブとして機能させ、新たな学術領域における国際的な地位を確立する。

Outline of Annual Research Achievements

令和3年度は以下の活動を行った。
1) 領域研究の推進:2021年11月16日、17日に領域会議・総括班会議(クローズド形式)をZoomとoViceにて開催した。領域会議には100名以上の参加があり、活発な議論が行われた。また、領域ホームページで紹介した優れた論文業績の第一著者が発表する「全能性領域・論文徹底解説シリーズ」を開始しした。11回にわたり、研究内容・成果のみならず、論文掲載までの苦労などが発表され、好評を博した。また、海外(英国、ドイツ、米国)の優れた研究者のセミナーも3回開催した。また、共催セミナーも行った。2) 領域ホームページを通じて、領域の成果をリアルタイムに領域内外の研究者へ広報した。令和3年度には、研究班員の責任著者によるScience, Nature Genetics, Nature Communications, PNAS, Nature Cell Biology等に掲載された10報の優れた論文紹介を行うことができた。3)若手研究者の育成のために、若手のみによるweb セミナーを全15回、毎週2名ずつ発表するwebinarシリーズを開催した。また、4月28日に、「非ゲノム情報複製機構」「全能性プログラム」合同若手勉強会を開催した。コロナ禍の影響により、急遽、webのみの開催としたが、約100名の参加者により、有意義な討論を行うことができた。4)研究支援班では、領域内における遺伝子改変マウスの作製支援、エピゲノム解析支援、顕微授精・クローン作製支援などの研究支援を行った。5) 公募研究の決定:令和4年2月28日に新学術領域研究専門委員会が行われ、審査員8名(計画班員3名および外部委員5名)による公募研究の方針について協議し決定を行った。16名の候補者を決定した。6)中間評価報告書を提出し、ヒアリング(オンライン)による報告を行った。

2: Research has progressed on the whole more than it was originally planned.

Reason

本研究は当初の計画に沿って順調に進展している。A01およびA02いずれも全能性の理解および制御につながる新しい研究成果を得ることができた。特に、研究項目や計画・公募研究の枠を越えた活発な共同研究が領域の推進力となっている。次世代を担う計画班若手5人(宮本、井上、新冨、石内、岡江)全員が責任あるいは筆頭著者として顕著な業績を挙げたことも特筆される。中間評価報告書作成時点で、英文原著論文 192件、総説16件、招待講演 国内88件、国際28件を数えた。また、ホームページでも解説記事を掲載した顕著な成果(IF10前後以上目安、班員が責任著者)は、29報にのぼった。新型コロナの影響を受けつつも、公開シンポジウムを2回、領域内シンポジウム(オンライン)を1回、若手勉強会を2回、論文徹底解説シリーズを11回、その他多くの海外研究者のセミナー、学会シンポジウムの共催等を行うことができた。総括班による技術支援も、遺伝子改変マウスの作製支援、エピゲノム解析支援、顕微授精・クローン作製支援が順調に実施され、領域内共同研究を協力に推進した。また、令和4年度11月に開催する国際シンポジウム(佐々木裕之代表特別推進研究、「配偶子インテグリティ」領域との3者合同)の準備を開始した。以上の様に、総じて、研究活動、成果発表、領域内共同研究・技術支援、若手育成が順調に進展していると言える。

Strategy for Future Research Activity

領域活動の発展には、総括班の強力な支援が必須である。これまでの総括班活動をさらに強力に進める。研究技術支援として、これまでのゲノム編集マウス作製、発生工学、次世代シーケンサー解析に加えて、需要が高まっている染色体 FISHと幹細胞樹立の支援を追加する。若手育成では、若手勉強会、若手webセミナー、海外学会参加をさらに充実し、自立的な研究の芽を育てるとともに、論文発表も目標とする基盤を醸成する。また、学会との共催シンポジウム、論文徹底解説シリーズ、海外研究者のセミナー、そして成果の公開も積極的に続けていく。なお、これまで困難に直面していたのが、国際活動である。コロナ感染の影響が続いても進められる班員の人的ネットワークを利用した国際共同研究支援、国際情報発信、国際web学会参加をサポートしていく。令和4年度は、佐々木裕之特別推進研究および「配偶子インテグリティ」領域との合同で国際公開シンポジウム(2022年11月23-25日、九大百年講堂)を開催する。多くの海外からの著名な講演者を招待し、多数の国際共同研究の端緒としたい。また、研究者の行き来が再開すれば、若手の国際学会への参加および海外研究室訪問をサポートして行きたい。もし、コロナ禍の影響が続く場合は、これまでと同様に、総括班予算を若手の支援、特に学生を含めオンライン国際学会や講習会の参加支援に充当する。
