Outline of Final Research Achievements

In this study, we identified a trans-omics network of metabolic adaptations and clarified the mechanisms of glucose metabolism in liver and muscle by omics measurement and data integration analysis of dynamic metabolic responses to glucose loading using ob/ob mice, a model of wild-type and type 2 diabetes mellitus. We have also developed OMELET, a mathematical model to analyze metabolic changes and their regulation based on multi-omics data without using isotope labeling experiments, and quantitatively clarified changes in liver glucose metabolism in ob/ob mice, a mouse model of obesity, and the regulation that causes these changes.

Academic Significance and Societal Importance of the Research Achievements

本研究では、野生型と肥満モデルマウスにおいてインスリンの標的臓器である肝臓や筋肉がいかにして代謝制御を行うかを初めて大規模に明らかにした。これにより代謝物による制御の消失や遅い遺伝子発現を介した制御など肥満における代謝制御の新たな側面が明らかとなった。血糖値の恒常性は肝臓のみならず全身の臓器連関ネットワークを介して維持されているため、他の代謝臓器にも本解析手法を拡張していくことにより、2型糖尿病など血糖恒常性の破綻メカニズムの全容が明らかとなると期待される。
