Outline of Research at the Start

精子および卵子ゲノムは、受精により全能性を獲得する。この全能性の獲得には、卵子細胞質が大きく関わる。体細胞核移植は、その卵子細胞質中に存在する母性因子を利用して体細胞ゲノムに全能性を賦与する技術であるが、核移植クローン胚の多くは発生停止をしたり、あるいは生後の異常表現型を呈する。よって、正常な受精および体細胞核移植クローンにより得られた胚・胎仔・産子を詳細に解析することにより、完全な全能性獲得のための条件が明らかになると期待される。本研究では、発生工学的技術と最新のゲノム解析技術を駆使することにより、全能性獲得とその後の正常な発生・出生・成育に至るエピゲノム条件を明らかにする。

Outline of Annual Research Achievements

本年度は、核移植クローンを通じた全能性エピゲノムの理解を目指し、以下の実験を行った。これまで多くの研究で、マウス体細胞核移植(SCNT)胚では、2細胞期における胚性遺伝子活性化(ZGA)の不全が報告され、その改善が進んでいるが、SCNT胚が発生停止する着床後の異常についての所見は少ない、そこで、SCNT胚の発生能の改善を目指して、桑実胚期に着目し、転移性エレメント発現異常の同定およびその機能解析を行なった。IVFおよびSCNT由来の桑実期胚を用いてsingle-embryo RNA-seqを実施し、転移性エレメントの発現を比較した。その結果、SCNT胚では、転移性エレメントの中でも特に内在性レトロウイルスに分類されるERVKの発現量がIVF胚の半分以下まで抑制されていた。着床前の受精胚では内在性レトロウイルスが高発現していることが報告されており、シス制御を介して遺伝子発現に何らかの影響を与える可能性がある。そこで、我々のRNA-seqデータと公開されているChIP-seqデータを組み合わせて、シス制御におけるERVKの役割を調べた。予想通り、ERVKのサブタイプRLTR45-intに隣接する遺伝子の発現量は、RLTR45-intの発現量に応じて変化していた。さらに、8細胞期IVF胚ではRLTR45-intがエンハンサーマーカーであるH3K27acに富むことが分かった。これらの結果から、少なくとも一部のERVKは桑実胚期に活性化する遺伝子のエンハンサーとして機能すること、そしてSCNT胚ではERVKの抑制によるそのエンハンサー活性の低下が着床前後の発生停止の一因である可能性が示唆された。現在、この ERVKのノックアウトおよびノックダウン実験を進めており、その着床後の胚発生に関わる機能を解明する予定である。

Reason

これまでに、クローン胚の発生に影響するエピジェネティクス異常としてH3K27me3の Loss of Imprintingによる Sfmbt2 miRNA の過剰発現（両アレル性発現）を同定し、その正常化によるクローン効率の向上を達成した。そして胎盤のエピゲノム特性をH3バリアントの観点から解明し、極めて強固なヘテロクロマチン領域が形成されていること、そしてそのヘテロクロマチンを弱めることで完全な全能性を回復し、クローンマウスが産まれることを証明した。今年度は着床直前のクローン胚におけるエンハンサー様ERVKの発現低下を明らかにした。この研究は、その後、ノックアウトあるはノックダウン実験につなげ、このERVKの役割を明らかにする。以上の成果は、今後の哺乳類のクローン技術の改善のみならず、本領域でも課題としている胎盤形成のエピジェネティクスの解明に貢献するものと期待される。よって、おおむね順調に進展していると考えられる。

Strategy for Future Research Activity

本年度までに、核移植クローンを通じた全能性エピゲノムの理解を目指し、ヒストンバリアントあるいはヒストン修飾の異常を明らかにし、その改善を行ってきた。H3K27me3依存性の胎盤特異的刷込み遺伝子の異常については、メジャーな遺伝子の解析は終了したが、その他の遺伝子の解析を進めている。また、これらの刷込み遺伝子のうちSfmbt2やGab1 などでは、発現アレルである父方アレルをKO すると、補償的に母方アレルが発現する現象がある。これは通常の刷込み遺伝子にはみられない現象であるが、本年度は本格的に進めることができなかったので、今後、そのメカニズムを明らかにしたい。ヒストンH3バリアントについては、胎盤系細胞におけるそのヘテロクロマチン形成への役割およびその胎盤系列維持機能を明らかにした。今後は、クローン初期胚におけるH3バリアントの解明を進めることにより、体細胞ゲノムの再プログラム化の詳細を解明する予定である。マウスクローンの大きな問題点である、胎仔無し胎盤の発生の原因は、刷込み遺伝子のIG-DMR 領域が関連していると考えているが、この問題は解決していない。現在、CATCH-seq という少数胚での解析を可能にする方法の導入を進めており、これが実用化した際には、IG-DMR 部位のヒストン修飾を解析する。、
