Outline of Research at the Start

細胞内寄生原虫であるトキソプラズマ原虫に対するセルオートノマス免疫系にIFN誘導性GTPaseの一つであるIRGB6が重要である。IRGB6は寄生胞膜に蓄積するPI5PをPLAMPとして認識する。しかし本来、健常な細胞(未感染細胞)では極めて低い濃度に保たれているPI5Pが、寄生胞膜上に蓄積するのかについてはよくわかっていない。さらに、PLAMPであるPI5Pが寄生胞膜にあるとIRGB6依存的に膜変化が起きる生化学的あるいは構造生物学的機序は全く分かっていない。そこで、本研究ではトキソプラズマ寄生胞膜上でPLAMPであるPI5Pが生成され、膜が破壊されるメカニズムを解明する。

Outline of Final Research Achievements

During the analysis of the host defense system against the pathogenic protozoan Toxoplasma, it was found that the cell-autonomous immune system, which determines self and non-self within cells, recognizes not pathogen-associated molecular patterns (PAMPs) but molecular patterns associated with the pathogen's life cycle, referred to as Pathogen Life-cycle Associated Molecular Patterns (PLAMPs). This PLAMP concept is novel compared to PAMPs, but empirical experiments are necessary to provide specificity to this concept. This study identified that the phospholipids on the parasitophorous vacuole membrane of Toxoplasma are the essence of PLAMPs, thereby concretizing the concept of PLAMPs.

Academic Significance and Societal Importance of the Research Achievements

本研究はIrgb6やIrgm2がPLAMP依存的なトキソプラズマ感染に対する宿主防御において重要な役割を果たしていることを明らかにしたことから、将来的な治療法の開発に寄与する医学的意義を有するものである。また、PLAMPを基盤とした様々なヒト用や家畜動物用のワクチンが開発されることも医学的な意義がある。さらにトキソプラズマは初感染の妊婦に感染し胎児・新生児が先天性トキソプラズマ症となったり、あるいは、大人でも免疫不全者で致死的な後天性トキソプラズマ症を引き起こすことから、これらの疾患を有する人々にとって新規の治療法・制御法につながる本研究は福音となる。
