Outline of Research at the Start

pHは生存に必須のパラメーターであり、その変化はストレスとなる一方で生命機能のシグナルとして働く。しかしその詳細な仕組みと意義は明らかではない。本研究では細胞質局所「pH場」の実態を発現系細胞やモデル生物のin vivo胚などにおいて詳細に明らかにするとともに、胚発生や神経機能や老化などのマクロな生物現象での生理的意義を解明することで、従来のホメオスタシス調節因子としての細胞内pHのコンセプトを超えたシグナル調節因子としての細胞内「pH場」の概念を確立する。

Outline of Final Research Achievements

We found cultured microglia has activities of Hv1 on endosomes which were revealed by patch clamp analysis and pharmacological experiments. These activities lead to regulation of actin polymerization through pH sensitive mediating proteins. We also found that killifish deficient of Hv1 gene has a longer lifespan, suggesting that local pH regulation by Hv1 might be involved in homeostasis of tissues. In our study of diapause of killifish, we found that intracellular pH shifts to an acidic state, and this acidification induces the diapause process.

Academic Significance and Societal Importance of the Research Achievements

pHは、細胞の恒常性や膜を介する電気化学勾配によるエネルギー産生を中心に研究されてきた。岡村らは貪食細胞の活性酸素産生時のpH制御因子であるHv1がミクログリア細胞内小胞上に発現し局所の「pH場」を形成し、細胞骨格の動態を調節することを見出した。また魚類の解析を通し発生や老化に伴うpH場の関与が示された。これらの成果は、pH場が発生や老化などの時系列現象の理解に繋がり、さらにこれらの変調による病態の解明や制御の戦略確立につながると期待される。また領域内連携によって有孔虫やサンゴのHv1オルソログ分子を明らかにし、今後生命環境丸ごとを盛り込んだpH場形成の仕組みと意義の解明へと進むと期待できる。
