Outline of Research at the Start

組織常在性記憶CD4+ T細胞が、肺の炎症記憶を制御し組織障害エントロピーの一端を担う細胞集団であることが示唆されるが、組織常在性記憶CD4+ T細胞の分化・維持に関する分子機構は、現在のところ全く不明である。そこで、本計画研究では、炎症記憶の制御機構を解明し、慢性気道炎症疾患の再燃や増悪のリスクを増加させる“組織障害エントロピー”の一端を明らかにするために、組織常在性記憶CD4+ T細胞の分化・維持機構に焦点をあて、1細胞レベル、クロマチンレベルおよび生体レベルの解析、患者での解析を進める。

Outline of Final Research Achievements

In order to elucidate the molecular mechanisms of inflammatory memory and to identify 'tissue damage entropy' that increase the risk of relapse or exacerbation of chronic airway inflammatory diseases, we analyzed the inflammatory memory at the single-cell, chromatin and biological levels, as well as in patients. We identified the mechanism of severe disease on viral infection and the mechanism of chronic inflammation pathogenesis associated with increased "tissue damage entropy" via the "neuro-immune interaction".

Academic Significance and Societal Importance of the Research Achievements

本研究の大きな成果の一つは、「免疫系」と「神経系」の相互連関によって形成される組織障害エントロピーに関して、その本質を細胞・分子・クロマチンレベルで明らかにしたことである。これは、今後の炎症収束学の創生へつながる学術的意義が大きい成果である。本研究領域の研究を通じてヒトの生体内において炎症記憶の制御機構を解き明かすことは、難治性疾患の病態制御へとつながり、社会貢献上のインパクトも大きい。
