Research Abstract

コレラの予防にはワクチンの経口投与が極めて有効である。従来、経口型のコレラワクチンとして、死菌ワクチンと組換え型の弱毒性菌ワクチンが検討されてきた。死菌ワクチンは運動性と粘着性を失活させてしまう加熱とホルマリン処理で作製されたもので、フィールド実験で高い安全性とある程度の感染防御効果が確認されている。本研究では、従来の死菌ワクチンがもつ高い安全性と生菌ワクチンがもつ高い有効性を兼ね備えた全く新しい概念のコレラ粘膜免疫ワクチンを開発することを目標とした。
新型コレラ菌0139ベンガル型をCFA寒天培地上で培養後、^<60>Co照射し、死菌体とした。10^9〜10^<10>(CFU)/mlレベルの菌液を用いた場合、1.5×10^5rad以上の^<60>Co照射が必要であった。^<60>Co照射後の死菌体は、6〜50%の残存HA活性を、43〜65%のレベルでM細胞への粘着能力を、5〜38%の運動性を依然保持していた。^<60>Co照射による殺菌のメカニズムは巨大構造であるDNAの破壊であった。以上の結果、新型コレラ菌0139を^<60>Co照射することによって、死んではいるが、依然粘着性と運動性を保持した状態の菌体を作製することができた。この菌体が示す粘着性はヒトパイエル板M細胞に対しても働くことを確認した。今後、この生菌ワクチンを用いて動物実験を行い、新しい経口型コレラ不活化ワクチンとしての有効性を確認する予定である。
