Research Abstract

1) Targeting vector was designed by inserting the exon4-5, including the phosphatase domain, between loxP sites to generate the conditional KO mice of PIEN.PTENflox/+ mice were obtained by injecting the PTENflox/+ ES cells into the blastocysts.
2) PTENflox/+ mice were crossed with Lck-Cre transgenic mice to delete PTEN gene in T cells. Ninty-five % of thymocytes were deleted PTEN at both DNA and protein level and this mutation is a null-mutation, confirming that Cre-loxP system worked well in vivo. All of the Lck-CrePTENflox/- mice developed into CD4 T cell lymphoma/leukemias in 17 weeks after birth.
3) To delete the PTEN gene specifically in brain, we are now crossing the PTENflox/+ mice with GFAP-Cre and MBP-Cre transgenic mice to generate GFAP-CrePTENflox/- and MBP-CrePTENflox/- mice.
4) PTEN+/- mice were observed to see the spectrum of suffering cancers. We found endometrial cancer and breast cancer were also seen in a longer observation period, although glioma has never been observed.
