Outline of Annual Research Achievements

筋萎縮側索硬化症(ALS)・前頭側頭型変性症(FTLD)患者の脳内に沈着したTDP-43凝集体に結合する化合物を見出すことをことができれば、それらをポジトロン放出核種で標識し、TDP-43病変を非侵襲的に計測することができる。TDP-43病変の非侵襲的な計測法は、アルツハイマー病で活用されているアミロイドイメージングのように、ALS・FTLDの早期診断や薬効評価系に有効活用することができると考えられる。今年度は、前年度剖検脳をスクリーニングして選定したTDP-43病変を高度に含むFTLD患者の脳切片を用いて研究室が保有する化合物ライブラリーを用いた蛍光染色およびI-125、ポジトロン標識(F-18)化合物を用いたIn vitro オートラジオグラフィーを実施した。
蛍光染色では、TDP-43病変を明瞭に染色される化合物は見いだせなかった。その一方で、I-125およびポジトロン標識(F-18)化合物を用いたIn vitro オートラジオグラフィーでは、TDP-43病変が高度に含まれているFTLD患者切片において集積を示す化合物を見いだすことができた。
また、その化合物の体内動態評価を行ったところ高い脳移行性を有し、その後のクリアランスも良好であった。この化合物はアミロイド/タウを含むAD脳切片においても集積を示し、今後さらなる最適化が必要である。
以上のことから、TDP-43病変に対して高い選択性は有してはいないが、オートラジオグラフィーで集積が認められるレベルの結合性を持つ化合物が見いだせた。今後は、この化合物をリード化合物としてさらなる最適化を進めることで、TDP-43病変の非侵襲的な計測系の開発を目指す。
