Outline of Final Research Achievements

Infertility is a global issue. To address it, I focused on finding and analyzing genes essential for male fertility using genetically modified (GM) animals. In this study, I generated CRISPR/Cas9-mediated GM mice and found several essential factors regulating the sperm fertilizing ability. In addition, I have established an efficient CRISPR/Cas9-mediated method to introduce point mutations and knock-ins that were previously produced at low efficiency. These genome-editing technologies are quite useful to make model animals of human genetic diseases.

Academic Significance and Societal Importance of the Research Achievements

本研究では、新たなゲノム編集技術CRISPR/Cas9を駆使して遺伝子組換えマウスを効率良く作製・解析することで、哺乳類の生殖に必須な遺伝子を複数同定することに成功した。その中でも特筆すべきは、研究代表者がこれまで着目してきた生殖細胞特異的に発現する膜タンパク質の一種GPIアンカータンパク質を網羅的に解析した結果、LYPD4が精子受精能に関与する新たな因子であることを明らかにした。今回発見した因子の全てがヒトにも保存されていることから、男性不妊の診断マーカーや避妊薬開発のターゲットとしての可能性を秘めている。
