Outline of Final Research Achievements |
Iron-sulfur (Fe-S) clusters are protein co-factor that are known to have extremely oxygen-labile nature. To elucidate the molecular mechanism involved in the Fe-S cluster biogenesis, in this proposal we focus on its core protein complex. We have been determined the crystal structure of the core complex, thereby the internal cavities were found. The mutagenesis in vivo assay and structural analyses implied that this cavity is probably play a role for the reservoir of iron and/or sulfur species for the de novo Fe-S cluster biosynthesis.

Academic Significance and Societal Importance of the Research Achievements |
鉄硫黄クラスターは、動物では呼吸、植物では光合成の中心を担う成分である。生物界において必須な役割を担う成分にも関わらず、いまだにこの生合成機構が明らかになっていない。この主な原因として、鉄硫黄クラスター自体が酸素に極端に不安定で、実験室では数秒~数分で崩壊してしまうことが挙げられる。本課題では、鉄硫黄クラスターの崩壊を防ぎながら、さまざまな実験方法を遂行し、これまでに分かっていなかったクラスター合成の材料の供給経路を明らかにできた。将来的にこれらの成果は、人間に副作用・悪影響がない抗生物質や除草剤の開発に繋がることが期待されている。
