Outline of Final Research Achievements

We explored novel Mib1 substrates for cell migration and found that Mib1 ubiquitinates p120ctn. Mib1-mediated ubiquitination of p120ctn K547 attenuated Rac1 activation in cultured cells. In addition, we found that posterior lateral line primordium cells in the zebrafish mib mutant showed increased random migration. Knockdown of p120ctn partially rescued posterior lateral line primordium cell migration defects in the mib mutant. Taken together, our data suggest that Mib plays an important role in cell migration.

Academic Significance and Societal Importance of the Research Achievements

細胞移動は動物の発生時や創傷治癒時などにみられる普遍的な生命現象だが、どのような分子が細胞移動を制御しているかは完全にはわかっていない。我々はMibと呼ばれるタンパク質がユビキチン化(ターゲットのタンパク質にユビキチンという別のタンパク質を付加すること)によって細胞移動の制御に関わるという知見を新たに見出した。今回の成果は動物の形づくりの理解やがん転移などの細胞運動が深くかかわる疾病研究に貢献すると期待される。
