Outline of Annual Research Achievements

最近,骨格筋から分泌される内分泌因子:マイオカインが複数同定されており,自己作用だけでなく,脂肪細胞や骨細胞とクロストーク(相互作用)することが報告されている。しかしながら,運動や食品摂取によって誘発されるマイオカインが動脈硬化改善に関与するか否かの検討は国内外を見ても皆無である。本研究は,動脈硬化に対する運動・食品摂取効果のメカニズムに関与するマイオカインを同定し,マイオカイン作用の貢献度や分子機序の解明から効率的な運動・食事プログラムの構築を目指すことを目的とする。本研究では,中高齢者(60 歳以上)45名を対象に安静対照群、有酸素性運動(週3回,1回45分間,60-70%VO2max運動強度の自転車エルゴメータ運動)群、アミノ酸摂取(必須アミノ酸摂取)群のランダムに3群に分けた。8-12週間の介入前後に,動脈硬化指標である、全身性(baPWV)および中心(cfPWV)動脈の脈波伝搬速度を測定し、同時に採血を行った。有酸素性トレーニングにより、VO2peakは有意に増大し、腹部内臓脂肪面積およびbaPWVおよびcfPWVは有意に低下した(P<0.05)。アミノ酸摂取ではbaPWVにおいてのみ有意に低下した(P<0.05)。各被験者の血液を用いて、すでに報告されているマイオカインや新規マイオカイン候補物質の血中レベルを測定し、動脈硬化に対する運動効果に関与するホルモン・ペプチドを複数検出することができた。さらに、栄養摂取による効果に関わるホルモン・タンパクに関してもいくつか検出することができているため、今後さらなる解析により効果関連候補バイオマーカーを同定する。

Reason

有酸素性トレーニングおよびアミノ酸摂取によって低下した動脈硬化度とともに変化したマイオカインについては複数同定できたが、これらのマイオカインが骨格筋から主に分泌されているのかを遺伝子発現レベルで確認した上で,今後,運動によるこれらのマイオカイン変動が動脈硬化の低下に対して関与しているのかについて動物実験を用いて検討する必要性がある。

Strategy for Future Research Activity

運動により血中濃度が増大したマイオカインが動脈硬化度の低下に対して貢献するのかを動物実験を用いて確認し,さらに,動脈血管組織においてどのような分子機序で動脈硬化を低下させるのかについて検討する。
