Outline of Final Research Achievements

In this research, I tried to clarify the regulatory mechanisms for trophoblast cell proliferation and differentiation using placental hypoplasia (= poor development) mouse models generated by somatic cell nuclear transfer. For this purpose, I generated the chimeric embryos by injection of embryonic stem cells from normal mice into blastocysts of the mouse model. The experimental results indicated that fetal (exogenous) factors might be responsible for the placental hypoplasia of cloned mouse embryos. In addition, I developed the technology associated with oocyte artificial activation necessary to generate cloned mouse embryos (Miura et al., J Reprod Dev, 2018).

申請者は本研究において、クローン胚盤胞に正常なES細胞を注入することで、「正常なES細胞由来の胎仔」と「クローン胚の胎盤」から構成されるキメラ胚を作製することに成功した。キメラ胚ではクローン胚で見られる組織学的異常が改善する傾向が見られたことから、クローン胚の胎盤異常が「胎仔性因子」に起因している可能性が示唆された。また、マウス卵の人為的な活性化に関連する技術の開発も行い、卵内のタンパク質量の制御や生殖補助医療への応用の可能性を示した。
