Staphylococcus aureus strain RN4220 without a drug resistance mechanism was transformed with aac(6′)-Ie/aph(2″)-Ia containing a promoter region using pND50 (E. coli-S. aureus shuttle cloning vector) to establish a bifunctional enzyme expressing strain. In addition, a transformant of RN4220 strain with pND50 vector was also established as a control strain for assay system. Two transformants were used to establish a system for the screening of active substances to overcome ABK resistance; approximately 5,000 samples of microbial cultures were screened and searched for active substances against the bifunctional enzyme expressing transformant in 3 years.
