Outline of Final Research Achievements

We carried out 4C-seq analysis of chicken DT40 cells. 4C-seq analysis showed some features of centromere region in interphase nuclei. In addition, we identified some factors that are involved in the centromere structure by using neocentromere forming cells and Auxin inducible Degron system in chicken DT40 cells.

Academic Significance and Societal Importance of the Research Achievements

遺伝情報を担う染色体の正確な分配は、細胞増殖に不可欠な現象であり、生命の基本原理とも言える。染色体分配の要となるキネトコア(動原体)はタンパク質の巨大複合体として染色体上のセントロメアDNA上に形成される。一方、セントロメア領域は、分裂期のみならず間期の核内でも特殊な構造と機能を有すると長く考えられてきたが、解析の困難さから間期のセントロメアの構造に関する知見は乏しかった。申請者はニワトリの細胞を用いることによって、間期核内におけるセントロメアの特殊な構造を検出することに成功した。この発見は間期細胞におけるセントロメアの役割解明に非常に重要なステップとなると期待される。
