Outline of Final Research Achievements

Serum proteome analysis is difficult to observe low concentrations of inflammation related proteins such as cytokines due to the very wide dynamic range of protein concentrations. In this study, I constructed a system to enrich inflammation-related proteins that bind to inflammatory cytokine receptors and analyze them using a highly sensitive liquid chromatography-mass spectrometry. Anti-CRP positive sera were applied to this analysis system and succeeded in discovering novel protein candidates related to inflammation.

Academic Significance and Societal Importance of the Research Achievements

生活習慣病や癌など様々な疾患と慢性炎症の関連が着目されている。また、炎症メカニズムは複雑で未だに原因不明な炎症疾患が存在する。そのため、炎症に関連したタンパク質を包括的に解析する技術、さらには未知の炎症関連タンパク質を探索できる技術が求められている。そこで、本研究では炎症受容体を用いて血中から炎症関連タンパク質を分析できるシステムを新たに構築した。その分析系を用いることによって、包括的に炎症関連タンパク質するだけでなく、新たな炎症関連タンパク質候補を見出すことに成功した。
