Outline of Final Research Achievements

We have established surgically induced cleft lip-like defect in a C57BL/6 strain fetus at the embryonic day 16.5 which achieving high reproducibility and histologically similar to the spontaneous cleft lip fetus.
We have found that to induce defect at least 1mm width × 2mm length upper lip defect must be needed.
Also, we have tried to transplant three-dimensional regenerative tissue originated from mesenchymal stem cell from mouse amniotic fluid.

Academic Significance and Societal Importance of the Research Achievements

口唇裂の新規治療法として胎児治療法が現実味を帯びつつあるが、臨床応用の前段階として実験動物における種々の条件検討が必要である。これまで山羊や霊長類等の妊娠期間の長い大型哺乳類においては外科的口唇裂モデルが報告されているが、マウスではない。マウスはヒトと遺伝子の相同性が高く、遺伝的背景が詳細に研究されており、入手しやすいため、さらなる研究の促進に有用である。
ヒトにおける口唇裂の胎児治療が実現すれば、口輪筋の再建により、自然な咀嚼・嚥下機能を獲得することが出来る。既存の標準的治療法から口唇形成術を省略することで、生涯消えない瘢痕の醜形を大幅に軽減し、患者を精神的苦痛から解放することができる。
