Outline of Final Research Achievements |
In this study, we sought to elucidate early sex-differentiation mechanism mediated by estrogen using amberjack, in which sex is determined by estrogen. By means of histological observations and massively parallel mRNA-seq, we succeeded to narrow down sex-determining period within a week. Differential expressed gene (DEG) analysis identified several DEGs in this period. These information will be valuable to further reveal estrogen-driven sex determination mechanism in amberjack and other teleosts.

我々はこれまでに、エストロゲンが性を直接決定できることが明らかにした。一方、なぜエストロゲンが性を決める(すなわち生殖腺の運命を決定する)ことができるのかは、依然として明らかになっていない。本研究ではその原因が、性決定期が分子レベルで特定されていないことにあると考え、性的に未分化な時期から性が確実に決まっている時期まで、生殖腺の遺伝子発現変化を経時的に観察することを試みた。 その結果、分子的な性分化が始まる時期を1週間程度まで狭めること、その時期の発現変動遺伝子を複数同定すること、に成功した。これらの遺伝子を詳細に解析すれば、エストロゲンによる性分化メカニズムの一端が明らかになると期待される。
