| Outline of Final Research Achievements |
IL-18 potentiates antitumor activity of immune-checkpoint blockade. In this study, we found followings; 1) NK cells recruited cDC1 cells into tumor sites in mice treated with IL-18 + anti-PD-1 mAb. 2) Accumulation of tumor neoantigen appeared crucial to overcome resistance to immunotherapy. 3) IL-18 reduced autoimmune liver toxicity while maintaining efficacy of tumor immunotherapy.

| Academic Significance and Societal Importance of the Research Achievements |
免疫チェックポイント阻害薬は担癌宿主のT細胞を抑制状態から解放し、強い抗腫瘍効果を発揮する。しかし、半数以上の患者には未だ十分な腫瘍縮小効果が見られない。また、克服すべき副作用として免疫関連有害事象が残されている。現在、免疫チェックポイント阻害薬の抗腫瘍効果を増強し、治療抵抗性を克服する新しい戦略の開発が強く望まれている。本邦で発見されたIL-18を、癌免疫応答を増強する医薬品として実用化することの臨床医学的な意義はきわめて大きい。
