Outline of Final Research Achievements

S100 proteins are highly expressed in CD274low cancer stem cell (CSC)-like cells in cholangiocarcinoma. Here we demonstrated S100A10 is indispensable for the tumorigenicity of the CSC-like cells. We also found the indispensable roles of S100A10 on cell cycle progression and cell migration of cholangiocarcinoma cells. S100A10 regulates the integrity of cytoskeletons, including vimentin intermediate filaments.

Academic Significance and Societal Importance of the Research Achievements

胆管癌幹細胞を含む細胞分画の指標である「CD274low」とはあくまで細胞の表現型であり、癌幹細胞としての機能発現への関わりを示すものではなかった。我々は、本細胞分画に高発現する分子としてS100分子群を同定し、特にS100A10に関する解析から癌幹細胞性維持における重要性を明らかにした。また、この分子が癌細胞の細胞遊走能獲得に関与することを示した。本研究の成果は、S100A10が発癌から転移まで広範囲に機能することを示唆しており新しい治療標的となりうる分子を見出した点で意義深いと考えている。
