Outline of Final Research Achievements

Ferroptosis is a newly found regulated necrosis and known as iron-dependent cell death. According to Dixon et al. (2013), lipid peroxidation plays an important role in cell membrane damage induced by ferroptosis instead of apoptosis. Ferrostatin-1 is a well-known inhibitor of cell death in the ferroptosis pathway of cancer cells. The erastin was also used in MC3T3E1 osteoblasts to influence erastin-induced cell death in cell differentiation tests. In addition, previously we found that ferrostatin-1 has a doping effect on osteoblast viability.

Academic Significance and Societal Importance of the Research Achievements

フェロトーシスは近年その存在が認められた新たな細胞死の概念であり、現在ではがん抑制メカニズムとして注目されている。これはがん細胞の酸化ストレスに対する抵抗性を減弱させ、 フェロトーシスという鉄依存的な lipid ROS の蓄積による細胞死を起こしやすくしている一方で、骨再生におけるフェロトーシスおよび lipid ROS の影響について報告した研究はなく、本研究は骨芽細胞への影響およびメカニズムを新たに解明し、それを阻害する基質を検索し、骨芽細胞死を減少させ、骨形成を促進する基質を見いだせたので、学術的・社会的意義が大きい。
