Outline of Final Research Achievements

DNA methylation is considered to be an important epigenetic modification for controlling gene expression. Since the discovery of the active DNA demethylation pathway in mammals, numerous efforts have been made to distinguish epigenetic cytosine variants, including 5-methylcytosine (5mC), 5-hydroxymethylcytosine (5hmC), 5-formylcytosine (5fC), and 5-carboxylcytosine (5caC). In this project, we developed a technique for the sequential multiple assessment of cytosine variants by a surface-plasmon-resonance (SPR)-based immunochemical assay.

Academic Significance and Societal Importance of the Research Achievements

ELISA法などの従来のイムノアッセイでは、一つのDNAに含まれる2種類以上の標的を評価できないため、4種のシトシンバリアントを一度に識別することが困難であった。本研究で開発したSPRイムノアッセイでは、同一DNAに含まれる4種のシトシンバリアントを連続して検出することに成功した。本研究で開発した手法は、シトシンバリアントの生物学的意義や産業的価値を明らかにする有用なツールになると期待できる。
