Research Abstract

土壌を接種源とする集積培養物TUT2264は、テトラクロロエテン(PCE)をジクロロエテン(DCEs)にまで脱塩素化する能力を発揮した。16S rRNA遺伝子を標的とした群集構造解析では、"Dehalococcoides"とAcidaminobacter属細菌の存在が確認された。また8種のRdhAsが見出され、転写レベルからもPCEおよびトリクロロエテンの脱塩素化が確認された一方で、分解活性が見られないDCEsおよびビニルクロライドによって転写が促進されていることが示された。クエン酸およびグルタミン酸がPCEの脱塩素化にとって有効な電子供与体となり得ることが示された。ヘッドスペース中の初期水素濃度(0%(v/v)と80%(v/v)がPCE脱塩素化に及ぼす影響を解析した結果、水素生産細菌のAcidaminobacter属細菌菌密度に差異は見られなかったが、"Dehalococcoides"においては0%の方が約100倍高く、分解活性も3倍程高かった。以上の結果は、異属微生物間の水素伝達系を良好に制御することで効果的な脱塩素化が可能なことを示唆している。
