Research Abstract

高酸素投与によるマウスARDSモデルをPYK2欠損マウスに作成したところ、野生型と比べ肺障害の著しい抑制があった。そのメカニズムとして、PYK2欠損により、NADPH-Oxidase由来の活性酸素の発現の抑制、サイトカイン産生の抑制による炎症反応の軽減が見られた。さらにアデノウイルスベクターを利用してPYK2-SiRNAを作成し野生型マウスに経気道的に投与したところ、肺障害の抑制が得られた。
