Research Abstract

われわれは、14q32領域に存在する刷り込み遺伝子GTL2遺伝子と同じであり、GTL2と同様epigeneticsで制御を受けている可能性が示唆されたmir-127について重点的に解析をおこなった。すなわちsiRNA によるノックダウンシステムを腎癌細胞株において作成し得られたWild株とノックダウン株についてマイクロアレイを行い、さらにその結果をRT-PCRおよびノーザンブロットでの確認を行った。この結果14q32領域の5つのmiR(miR-433,miR-127,miR-299,miR-134, miR-154)の発現が腎癌細胞株で高頻度に消失している(83%)ことを明らかとした。しかしながら、腎癌発生に寄与するmiRとその標的遺伝子の同定にまでは至っておらず今後の検討が必要とされる。また,miR発現を制御する機構についてもDNAメチル化だけではなくヒストンのメチル化・アセチル化の検討が必要と考えられた。
