Outline of Research at the Start

好塩基球は末梢血白血球のわずか0.5%を占めるに過ぎない希少細胞で、末梢組織常在のマスト(肥満)細胞と類似の特徴を有するため、生体内での存在意義が長い間疑問視されていた。最近、私たちは好塩基球が寄生虫感染防御やアレルギー炎症反応においてマスト細胞とは異なる固有かつ重要な役割を果たしていることを明らかにした。本研究では、独自に開発したツールを駆使して生理的条件下ならびに疾患病態における好塩基球の動態、活性化、エフェクター機能に関わる分子機構を明らかにし、それに基づき好塩基球ならびにその産物を標的とした炎症性疾患や感染症の人為的制御の可能性を探求する。

Outline of Final Research Achievements

Basophils are the rarest granulocytes circulating in the peripheral blood and evolutionally conserved in many animal species. Their functional significance remained an enigma for a long time because the basophil research has been hampered by their paucity. In this study, we took advantage of newly-developed analytical tools and have clarified that basophils play crucial roles in vivo, either beneficial or deleterious ones, depending on health and disease conditions.

Academic Significance and Societal Importance of the Research Achievements

好塩基球は希少細胞である上に末梢組織常在のマスト細胞と一部類似する特徴を有することから、生体内での存在意義が長い間疑問視されてきた。本研究により、好塩基球はマスト細胞とは異なる固有な働きをしており、数こそ少ないものの、病気の発症や回復に寄与していることが明らかになった。この研究成果を踏まえてヒトの好塩基球研究をさらに進めることで、好塩基球を標的にした病気の新たな制御方法の開発が期待される。
