Outline of Research at the Start

選択的ミトコンドリア分解は酵母からヒトまで保存されたミトコンドリア質・量管理システムの一つである。本研究の目的は、選択的ミトコンドリア分解の制御機構を分子レベルで理解することであり、とりわけ、分解の初期段階における誘導機構や分解効率・選択性を調節する基本原理の理解を目指す。具体的には、①分解に重要なミトコンドリア断片化を司るタンパク質の機能、②栄養/ストレスシグナリング経路を介した分解の誘導、③分解に必須な分別標識タンパク質の局在と発現量の制御、④小胞体膜タンパク質生合成システムによる分解の促進、の4項目を明らかにしてゆく。

Outline of Final Research Achievements

Selective sequestration and removal of mitochondria as a whole organelle is a fundamental mechanism conserved beyond species. Defects in this autophagy-dependent process, termed mitophagy, are associated with various pathologies, highlighting its biological relevance. However, detailed molecular mechanisms underlying mitophagy remain elusive. In this study, we sought to investigate how Get1/2, a complex promoting insertion of membrane proteins into the endoplasmic reticulum, acts in budding yeast mitophagy. We found that Get1/2 contributes to mitophagy via localizing Ppg1-Far, a factor negatively regulating mitophagy, to the endoplasmic reticulum.

Academic Significance and Societal Importance of the Research Achievements

これまでの研究から、マイトファジーが駆動される際、オートファジーの仕組みをミトコンドリアへリクルートするための「分解標識」タンパク質が活性化する。このマイトファジー駆動タンパク質を負に制御する因子が小胞体に局在していること、その一部がミトコンドリアへ標的化して、マイトファジー駆動タンパク質を抑制していることが明らかとなった。重要なことに、この負の制御因子の小胞体局在に働くタンパク質がマイトファジーに寄与している。このことは、選択的ミトコンドリア分解を司る仕組みが別のオルガネラにも存在していることを示しており、当該分野に新たな視点を与えるものである。
