Outline of Research at the Start

ミトコンドリア（mt）の細菌起源を鑑みると、そのゲノムはかつて細菌型DNA polにより複製されていたはずである。一部の真核系統ではミトコンドリアDNA pol（mtDNA pol）が同定されているが、既知のmtDNA polは細菌型DNA polとは異なる。従って、真核生物が多様化する過程で細菌型DNA polから進化的に異なるDNA polへの変換がおこったことを示唆するが、その詳細は未解明である。本研究では(1)多様な真核系統におけるmtDNA pol候補の探索と(2)その細胞内局在の確認を行い、真核生物の多様化に伴うmtDNA pol多様化と進化の全容解明を目指す。

Outline of Annual Research Achievements

これまでMarec Elias博士（オストラバ大学・チェコ共和国）とEnsoo Kim博士（梨花女子大学・韓国）と共同で行ってきた広範な真核生物におけるオルガネラ局在DNAポリメラーゼの多様性と進化についての研究成果を取り纏めた。まず2023年7月9-14日に、ウィーン大学（オーストリア）で開催されたIX ECOP-ISOP joint meeting 2023と3rd International Congress of Euglenoids 2023において稲垣の指導学生が本研究の成果を口頭発表した。特に前者の学会では、指導学生はトラベルアワードに選出された。また並行して研究成果を査読付き英文論文2報、邦文総説（査読無）1報として纏めた。特に英文論文1報は進化生物学分野におけるトップジャーナルであるMolecular Biology and Evolutoin誌（最新インパクトファクターは10.2）に掲載された。さらに、緑藻類で発見された2種類のDNAポリメラーゼ（chloroPolA & pyramiPolA）の細胞内の局在を実験的に検証しようと試みている。2種類のDNAポリメラーゼのN末端配列を蛍光タンパク質と融合し、その融合タンパク質をモデル生物である緑藻クラミドモナスの細胞内で発現させる予定である。この実験が完了した暁には、今後上記2種のDNAポリメラーゼについて緑藻細胞内局在を解明できる。また、いまのところ進化的起源が不明のままであるDNAポリメラーゼの1つPOPについて、公共データベースにデポジットされたメタゲノムデータ中のDNAポリメラーゼを探索し、POP配列に強い近縁性のある細菌とファージDNAポリメラーゼを探索している。
