Outline of Research at the Start |
先天性骨格筋欠損に対しての骨格筋シートの作成を最終目標とする.線維芽細胞を高効率に多核筋細胞に形質転換するダイレクトリプログラミング法と生体内組織形成技術により作成したバイオシートを用いることで,マウス腹壁欠損モデルについて,自己細胞・組織から強度と運動性を担保した「機能する」骨格筋シートを短期間に作成し,実臨床において応用可能な治療プラットフォームの開発を目指す.

Outline of Final Research Achievements |
In order to create skeletal muscle sheets for congenital skeletal muscle defects, we have developed a short time frame to create "functional" skeletal muscle sheets in a mouse abdominal wall defect model by using a direct programming method that efficiently transforms fibroblasts into multinuclear muscle cells and biosheets created by in vivo tissue formation technology. Specifically, multinucleated skeletal muscle cells differentiated from human fibroblasts by direct conversion were seeded onto a mouse abdominal wall defect model using a biosheet fabricated subcutaneously in mice as a scaffold, and transplanted into the mouse abdominal wall defect model for histological evaluation. The results showed some growth, but not enough. In vivo transplantation experiments using cross-linked nanogels will be performed in the future.

本研究では,先天性骨格筋欠損に対しての骨格筋シートの作成を最終目標とする.線維芽細胞を高効率に多核筋細胞に形質転換するダイレクトリプログラミング法と生体内組織形成技術により作成したバイオシートを用いることで,マウス腹壁欠損モデルについて,自己細胞・組織から強度と運動性を担保した「機能する」骨格筋シートを短期間に作成し,実臨床において応用可能な治療プラットフォームの開発を目指す.
