Outline of Research at the Start

申請者はこれまでに1) 治療抵抗性・食道扁平上皮癌・マイクロRNA発現プロファイルを作成し、治療抵抗性を示した患者血液中に存在するエクソソームから、2) エクソソーム由来・治療抵抗性・食道扁平上皮癌・マイクロRNA発現プロファイルを作成した。本研究で、上記プロファイルに基づき、食道扁平上皮癌の治療抵抗性に関わる分子機序を明らかにする。ゲノム編集を行った癌細胞から、食道癌オルガノイドを作成し、抗癌剤の感受性試験を行う。治療抵抗性を獲得した食道扁平上皮癌細胞において、活性化している分子経路を遮断する戦略の考案(ドラッグ・リポジション)は、本疾患の新規治療法の開発に向け有用な情報を提供可能と考える。

Outline of Final Research Achievements

Using the microRNA expression profile in Esophageal squamous cell carcinoma (ESCC), it was demonstrated that miR-143-5p, miR-143-3p, function as ESCC-suppressing microRNA.
HMGA2, KRT80 were found as new target genes, and their functional analysis was performed as esophageal squamous cell cancer genes. Furthermore, we demonstrated that these genes are associated with ESCC and play an important role in the functional molecular network of ESCC.

Academic Significance and Societal Importance of the Research Achievements

有効な治療法の少ない食道扁平上皮癌において、新規の食道扁平上皮癌マイクロRNAを解析することで、食道扁平上皮癌に関連するターゲット遺伝子を発見し、未だ報告のない新規分子やそれらの制御する機能性RNAネットワークの探索を行うことが可能だった。しかし、CRISPER-Cas9システムで癌促進型マイクロRNAのゲノム編集を行なった癌細胞の作成、食道癌オルガノイド作成については手技確率と報告に至らず、今後継続した研究が必要である。
