Outline of Research at the Start

近年、薬物有害反応(副作用)や薬物感受性(薬効)と薬剤標的分子の遺伝子変異との関連が明らかになりつつあり、例えば、肺がん分子標的薬は変異型上皮成長因子受容体(EGFR)の遺伝子検査結果に基づいて投与される。しかし、これまでに報告されたがん細胞でのEGFRの遺伝子変異では、一部を除きその薬物感受性は不明のままである。本研究では、肺がん分子標的薬と各変異型EGFRとの分子シミュレーション解析とその数理モデル化により薬効予測を可能にすることを目標とし、将来的には安全かつ効率的な薬剤投与の実現を目指す。

Outline of Final Research Achievements

Drug sensitivity (drug efficacy) is known to be associated with genetic mutations of drug target molecules. In this study, we used molecular simulation analysis to reproduce drug interactions between EGFR mutations and gefitinib, a molecular target drug for lung cancer (EGFR-TKI) that targets the epidermal growth factor receptor (EGFR), and elucidated the characteristics related to drug sensitivity of each mutant EGFR.

Academic Significance and Societal Importance of the Research Achievements

本研究の学術的意義は、肺がん細胞で新規変異体の発見が続くEGFRに対して抗がん剤を選択する可否の判定を実現する手段を提供する点である。東アジアにおけるEGFR変異の頻度は高く、科学的な効果予測因子として認識されている。また、本研究の解析方法は、あらゆる分子標的薬の評価や創薬にも応用可能である。
