Outline of Research at the Start

近年、エクソソームとよばれる顆粒がさまざまな生理作用を有することが明らかにされた。本研究では、血液や骨格筋の培養液などからエクソソームを単離・回収し、細胞内代謝環境の変化や筋収縮刺激に伴うエクソソームの量的・質的変動を明らかにし、運動に伴う骨格筋由来の生理活性因子による生体調節機構に新たな知見を得る。また、運動に伴うエクソソームの変動を検証し、運動やストレスを反映する新規バイオマーカーとしての妥当性を検討する。

Outline of Annual Research Achievements

運動に伴い骨格筋から分泌されるエクソソームの質的な変動とその役割についての検証を進めてきた。マウスの骨格筋に電気刺激を負荷する筋収縮モデルを用いて、血液中のエクソソームに含まれるマイクロ（mi）RNAおよびメッセンジャー（m）RNAの発現量の変化を次世代シーケンサーを用いて網羅的に解析した結果、筋収縮によりmiR-1やmiR-133などの骨格筋特異的miRNAの発現量が増加することを明らかにした。また、マウス胎児脳の海馬由来初代神経細胞にエクソソームを添加することによるmRNAの差次的発現解析を実施した結果として、筋収縮を負荷したマウスの血液中のエクソソームは132種類のmRNAの発現を変化させることを見出した。Gene Ontology解析によって発現変動したmRNAは細胞増殖などの機能活性があることが示された。
また、マウスから摘出した骨格筋を培養して、培養液中に分泌されるエクソソームの変動についても検証した。高脂肪食負荷による肥満モデルマウスに持久的走行運動トレーニングを負荷することで、miR-486bなどの複数のmiRNAの発現量に関して骨格筋が分泌したエクソソームで変化することを明らかにした。また、異なる時間帯での運動トレーニングの影響を検討した結果、明期と暗期の運動トレーニングではmiR-434などの複数のmiRNAの発現量に違いが認められた。さらに、肝臓や脂肪組織由来のエクソソームについても検討したほか、各組織からのエクソソームの抽出法と計測法を比較検討し、今後の実験での方法論についても知見を得ることができた。
運動と組み合わせる栄養素・機能性食品の効果と作用機序に関する情報収集と基礎的検討も進め、今後の応用的研究の足掛かりとすることができた。
