Outline of Research at the Start

本研究では、干し柿のタンニンが生体に対して抗酸化性を発現するプロセスについて、腸内細菌に着目して検討を行う。具体的には、干し柿に含まれるタンニンを分解して低分子化する能力を有する腸内細菌を探索するとともに、腸内細菌が生成するタンニン分解物の化学構造の解析および抗酸化性の評価を行う。さらにタンニンの投与が腸内細菌叢を含め生体に及ぼす影響についても評価を行う。これらの検討を通じて、干し柿に含まれるタンニンが有する抗酸化性と腸内細菌との関連、および生体に及ぼす健康増進効果を明らかにする。

Outline of Final Research Achievements

In this study, it was speculated that a species bacteria belonging to the phylum Bacteroidetes degrades tannins in the non-extractable fraction of dried persimmons (NEP) to produce low-molecular-weight antioxidants. Furthermore, administration of NEP to type I diabetic rats showed a significant increase in hepatic oxygen radical absorptive capacity (ORAC) and improvement of hepatic antioxidant enzymes such as superoxide dismutase, catalase, and glutathione peroxidase. A significant suppression of cross-sectional area reduction of extensor digitorum longus (EDL) muscle fibers was also observed. These results presume that the tannins contained in NEP ameliorate type I diabetic status.

Academic Significance and Societal Importance of the Research Achievements

これまでの研究において、植物性食品の機能性成分は主として可溶性画分に焦点があてられてきた。本研究では干し柿の不溶性画分に含まれるタンニンに着目し、機能性の発現に腸内細菌が関与する可能性を見いだすとともに、Ⅰ型糖尿病の病態を改善する効果を示す有益なデータを得た。これまでタンニンは防水剤、防腐剤、清酒製造の清澄剤などに利用されているが、食品成分として健康増進効果を評価した研究は極めて少ない。本研究の成果より、タンニンの機能性により注目が集まり研究が広がることによって、国民の健康増進に寄与することが期待される。
