Outline of Research at the Start

糖尿病治療をゴールとした多能性幹細胞からの膵β細胞分化誘導技術は着実に進歩を遂げてきてはいるが、未だに臨床応用に至っていない。つまりインスリンを発現するβ様細胞の創出はできるようになったが、十分な機能をもつβ細胞を作製する技術が未だに確立していない。それは、目指すべき機能的β細胞のprofileがヒトでは充分でなかった為に、作成した細胞がどこまで成体の機能的細胞に近似しているかの評価ができなかったからである。本研究では、申請者が明らかにしたヒト機能的β細胞プロファイルの「カタログ」を参照することで、作製過程および作製したβ細胞のインスリン分泌機構を多元的に評価して、分化誘導法を改良したい。

Outline of Final Research Achievements

Despite tremendous efforts since iPSC technologies were developed, we have not yet launched iPSC-derived β-cell therapy for diabetic patients.
In this project we established a novel protocol to differentiate human iPSCs into functional β-cells. By focusing on recapitulation of 3D-architecture of pancreatic islets and referring to the profiling of human primary β-cells, we improved our differentiation protocol, achieving more proliferation and better functionality in iPSC-derived β-cells.

Academic Significance and Societal Importance of the Research Achievements

iPS細胞の医療応用が様々な分野で推し進められているが、糖尿病治療を目指したiPS細胞由来の膵β細胞移植は、未だ実用化の目処がついていない。それは、インスリンを発現するβ様の細胞 (β-like cells)の創出は比較的容易にできるようになったが、十分な機能をもつβ細胞を再現性良く作製する技術が未だに確立できなかったからである。本研究では、これまでの研究で欠けていた視点を補い、新たな作製法を開発した。本研究成果が臨床応用への弾みとなるだけでなく、遺伝性糖尿病など疾患の解明にも貢献すると期待される。
