Outline of Research at the Start

Periodontal tissue is composed of cementum, periodontal ligament (PDL) and alveolar bone, given the complexity of this redundant structure, a single type of cell is not enough to regenerate such combined tissue. Here we are developing a complex cell sheet composed of PDL and osteogenic differentiated PDL cells. Furthermore, we will be analyzing the regenerative capacity of this cell sheet by ectopic and orthotopic transplantation in mice.

Outline of Annual Research Achievements

Complex cell sheet, CCS was made using rat PDL cells and osteogenic differentiated PDL cells. Transplants from ectopic transplantation analysis using histology and micro-CT showed the formation of periodontal ligament like bone like tissue. Furthermore, we have successfully created an orthotropic injury mice model for the transplantation of cell sheets. In a continuation study, we have evaluated the regenerative capacity of tooth dentin-pulp tissue by a CCS. Dental pulp stem cells, DPSCs and odontogenic differentiated DPSCs were utilized to fabricate a CCS. Ectopic transplants after 8 weeks of transplantation revealed the formation of dentin-pulp like combined hard and soft tissue formation.
