Outline of Research at the Start

応募者らが活性イオウ分子を発見して以来、生物(特に高等動物)におけるイオウの重要性が再認識されている。活性イオウ分子は、多様な存在様式を示すが、最近、応募者らは、タンパク質活性イオウ化が、翻訳共役型プロセスであり、すべてのシステイン含有タンパク質が活性イオウ化されていることを明らかにした。本研究では、応募者が明らかにしてきた活性イオウ分子の諸性質、開発してきた解析技術、試薬に加え、さらに活性イオウ化タンパク質の解析に特化した技術を開発し、活性イオウ化タンパク質の解析方法を確立することを目的とし、活性イオウプロテオミクスの基盤とする。

Outline of Final Research Achievements

To date, no method has been established for the analysis of reactive sulfuration (polysulfidation) of proteins. The aim of this study was to establish a method for analyzing polysulfidated proteins using LC-MS/MS.
We developed a method to analyze oxidized polysulfide structures in proteins using LC-MS/MS, which has been difficult to analyze so far, and showed that E. coli YgaV, a bacterial hydrogen sulfide sensor protein, forms intramolecular tetrasulfide structures in a hydrogen sulfide-dependent manner.
We also analyzed the intracellular reduced polysulfide sites of Keap1,an oxidative stress sensor protein, using LC-MS/MS and successfully detected 19 polysulfidated cysteine residues of 25 residues.

Academic Significance and Societal Importance of the Research Achievements

応募者らにより、すべてのシステイン含有タンパク質は活性イオウ化されていることが明らかにされたが、タンパク質中の活性イオウの存在様式、つまり「どのタンパク質の、どのシステインが、どのように活性イオウ化(還元型、酸化型、イオウの数)されているのか」は不明であった。
本研究により、活性イオウ化タンパク質の解析が可能となり、そこから得られる新たな知見が、タンパク質の機能制御、立体構造形成メカニズム解明の手掛かりとなり、タンパク質科学の新展開につながると期待される。
また本研究成果は、基礎生物学のみならず、医薬、薬学、農学、工学分野においても新領域を開拓するための礎になることが期待される。
