Outline of Research at the Start

北海道産ニンニクには2つのpotyvirus、ネギ萎縮ウイルス（OYDV）とリーキ黄色条斑ウイルス（LYSV）が混合感染している。OYDVのアブラムシ伝搬に必須のタンパク質HC-Proのシーケンスを解析したところ、N末端に大きな欠失があり、北海道産OYDVはアブラムシ伝搬能を失っているものと推測された。研究代表者は、OYDVはアブラムシ伝搬をLYSVに依存しているのではないかという仮説を持った。アブラムシがウイルスを獲得するには、CP-HC-Pro複合体がアブラムシの口針のStylin受容体に結合できるのではないかと想定され、この結合の存在の有無を明らかにする。

Outline of Annual Research Achievements

ポティウイルスのHC-Proは、アブラムシの口針にあるタンパク質（StylinやPRI）と結合できる他、ウイルスのコートタンパク質とも結合する。ニンニクには２つのポティウイルス、タマネギ萎縮ウイルス（OYDV）とリーキ黄色条斑ウイルス（LYSV）が混合感染して検出される。中国産や北海道産のOYDVはHC-Proのアブラムシ伝搬に重要なアミノ酸配列に長い欠損があるため、OYDVのアブラムシ伝搬は、LYSVのHC-Proに依存しているものと推定された。以前の研究で、短いHC-Proは、StylinやPRIと結合できないことをin vitroの結合試験で証明している。この短いHC-ProはRNA silencing suppressor (RSS)活性を喪失していない。しかも、中国産のHC-Proは長いHC-Proよりも高いRSS活性を有していた。HC-ProのRSS活性はウイルスの宿主適応に極めて重要である。ここで、我々は、OYDVがLYSVにアブラムシ伝搬を依存する現象は、OYDVの拡散・進化に本質的なことではないと考えた。そこで、バイオインフォマティクスとラボ実験を融合・駆使した研究手法をとりいれ解析した結果、ポティウイルスの拡散・宿主適応進化について明瞭な仮説を提唱することができた。すなわち、LYSVは進化の過程でP1タンパク質に生じた欠失によって、RSS活性を獲得し、HC-ProのRSS活性を強化することができたと考えた。この欠損P1をもつLYSVは、強化されたRSS活性によって宿主範囲を拡大させ、日本で優占種になったものと推定される。OYDVはLYSVのRSS活性を巧みに利用できたのではないか。あるいは、両ウイルスのHC-ProのRSS活性は相乗効果を発揮できた可能性すら考えられる。OYDVにとってアブラムシ伝搬能を喪失したことは、進化にむしろ有利に働いたと結論した。
