Outline of Research at the Start

植物根から影響を受ける根の近傍領域「根圏」では、植物と根圏微生物が密接な関係を構築し、ホロビオントとして機能する。本研究ではダイズ根圏ホロビオントを対象とし、植物と微生物の両方向から協奏的に、根圏の代謝物と微生物の時空間的変動を明らかにすることで、ホロビオントの形成過程と機能を分子レベルで解明する。

Outline of Annual Research Achievements

根圏とは「植物根から影響を受ける領域」と定義される根近傍の土壌領域である。根圏ではでは植物と根圏微生物叢がホロビオントとして一体的に機能すると考えられており、根圏ホロビオント形成に植物特化（二次）代謝産物が重要な役割を担うと考えられてきた。本研究では、植物特化代謝産物の根圏での動態と、それらを代謝する微生物の機能を時空間的に解析することにより、ホロビオント形成の分子機構と植物生育に及ぼす影響を明らかにすることを目的とした。今年度は、前年度までに絞り込んだ候補遺伝子の解析を進めるとともに、Variovorax属のパンゲノム解析を行った。イソフラボンを酸化的に異化する経路の主要な遺伝子群を同定したので、これらの遺伝子を含むクラスターをisoflavone catabolism gene clusterと命名し、ifc遺伝子クラスターを有するVariovorax菌がどのような分布になっているのかを調べた。その結果、ifc遺伝子を有する菌はVariovox属に広く存在するものの、イソフラボンを生産するマメ科植物の根圏から分離された菌株により多く見出された。このことから、ifc遺伝子クラスターを有することでマメ科植物の根圏への定着に有利である可能性が考えられる。さらに、ifc遺伝子を有するVariovox属細菌のイソフラボン存在条件での生育を詳細に解析し、Variovox属細菌がダイゼインを単一の炭素源として生育することができることや、イソフラボン存在条件下での生育抑制が低減されることを見出した。これらの結果について原著論文に取りまとめ発表した。
